PRESENTATION :
Each Cereton tablet contains Vinpocetine INN 5mg
Mechanism of action :
It has three ways to act
a. It increases cerebral metabolism by increasing glucose and oxygen
utilization.
b. It improves cerebral microcirculation by inhibiting platelet aggregation,
decreasing blood viscosity and promoting oxygen transport.
c. It increases cerebral blood flow.
Sodium channel blocking activity of CERETON :
There are a lot of Na+, K+, Ca++ in our body. Normally the concentration of
sodium ion in the cell is less than that of outside the cell. This ion can
penetrate into cell through cell membrane. In the normal condition of our body
control the concentration of Na+. When heat, cold, joy or sorrow disturbs the
normal or restoring condition, mental excitation and disease condition the
permeability of Na+ is increases. At that time Na+ enter into neuron cell, as a
result an electrical impulse is produced which is called neural convulsion. Due
to excess neural convulsion nerves cell can be affected.
By controlling the permeability of Na+ CERETON can prevent neural convulsions.
It is knwon as neuroprotective and anticonvulsant effect.
Five main pharmacological and biochemical actions of vinpocetin
1. Selective enhancement of the brain circulation and oxygen utilization without
alteration in parameters of systemic circulation.
2. Increased tolerance of brain toward hypoxia and ischaemia.
3. Anticonvulsant activity
4. Inhibitory effect on phosphodiesterase (PDE) enzyme &
5. Improvement of rheological properties of the blood and inhibition of
aggregation of thrombocytes.

Acute cerebro vascular accidents (Stroke) :
Ischaemic strokes due to cerebral
thrombosis, cerebral embolism, acute circulatory disorder, hypertensive crisis,
the acute cerebrovascular disorder, ischaemic neurological deficit (RIND),
complete stroke (CS), multi infarct dementia, cerebral arteriosclerosis,
hypertensive encephalopathy, Post apoplectic condition with the background of
haemorrhagic stroke etc.
Gerontology
For relief of psychosomatic
symptoms in the elderly due to cerebral insufficiency e.g. forgetfulness, memory
distrubances, slow thinking, lack of concentration, dizziness, mood instability,
aphasia, sleep disturbances, vasovagetative symptoms of menopausal syndrome etc.
Ophthalmology
Vascular disorders of the choroid
and retina due to arteriosclerosis vasospasm, macula degeneration, arterial or
venous thrombosis or embolism and glaucoma secondary to the above mentioned
disorder.
Otology
For the treatment of impaired
hearing vascular toxic (iatrogenic) origin, presbyacusis, meniere's disease,
cochleovestibular neuritis, tinnitus and dizziness of labyrinth origin.
DOSAGE
1-2 tablets thrice daily. The
maintenance dose is 1 tablet thrice daily.
PHARMACOKINETIC PARAMETERS
Vinpocetine is rapidly absorbed
after oral administration. Peak plasma concentration reached after 90 minutes.
Plasma protein binding is 66%. Kinetics of Vinpocetine in elderly does not difer
from that of the young. No need of dose reduction in case of disturbed liver or
kidney function.
